Analyzing Appearance of White Blood Cells on CellProfiler

Hi,

I am new to using Cell Profiler and I know that the ‘Identify Primary Objects’ Pipeline can be used for counting cells, etc. I have slides with samples of WBCs on them (primarily neutrophils) and want to identify cells with abnormal borders, multi-lobed nuclei, and hyper or hypo granulation. Is it possible to be this specific on CellProfiler and select abnormal cells? How would I go about it?

Hi,

You have to first detect nuclei and cells using identify Primary Objects and identify Secondary Objects modules respectively. Then you can extract size and shape features for detected cells and nuclei object intensity and Granularity of your images. After extracting these features you can use classifyObjects modules to classify cells based on single or pair of measurements in to different assigned bins according to the value of measurements such as by size, intensity, shape that were calculated earlier.
